Is there a way to increase font size of listed files and folders?

You mean:
Tools > Settings > Thumbnail > Appearance > Label Alligment
Tools > Settings > Browser > Thumbnail > Appearance > Label Alignment
?
If yes: then I have changed the font, clicked OK and re-opened the program - but I see no change

yes in Tools>Settings>Thumbnail>Appearance , you change the font & font size

Right, clicking on the displayed font opens a menu that enables
both the font and the font size to be set...

well, I would
- as minimum
- group label stuff
- add 'Font' label
- change font selection button to display font name only without size, style and effect as it cause some unpleasant UI behaviour
(do it for all Font buttons in Settings)

- rearrange elements a little bit on Appearance tab
- maybe change group naming from 'Thumbnail cell' to 'Thumbnail card'

I doubt separating font name and size would be good approach here, as these settings are not that important and you do not change them that often.
Also, in case of showing font name and size separately I would expect to have style (regular/bold/etc) and effect (strikeout,underline) controls to be present as well, which overwhelm UI too much.

Renaming the existing [Selected font name] button to Font would seem a simple start...
That would open the existing font menu with the typeface, size and style options, and should avoid the issue that the OP encountered.

Despite switching from using installed version 1.0.1 to portable 1.4.3 my choices for font are still being ignored

Yes
Also in version 1.0.1. And so as my font choice have been ignored it was my reasoning for investing time and energy in downloading 1.4.3
But the update have not fixed the issue

As you can see, chosen font is only applied to its button
And also:
Tools > Settings > Browser > Thumbnail > Appearance > Label Alignment > Font style
uses wrong language, as it is not English - i.e. the language I have chosen for my Windows 10 Enterprise 20H2 19042 x64 and for my XnView. That other one I have only under the
Menu Start > Settings > Time & Language > Preferred Language
of Windows set as a second keyboard of OS
